Endospores of Bacillus subtilis with probiotic potential in animals of zootechnical interest

At present, poultry, pig and cattle production constitute the most important branches of animal production in the world, which currently contributes to fulfill the protein needs of the world population. The use of probiotics, prebiotics and synbiotics is predicted with the purpose of using them as animal growth promoters. Among the species that are most used for the elaboration of these products, are those from Bacillus genus. One of them is Bacillus subtilis, a bacterium able to stimulate the immune system, produce enzymes, resist acid pH gastric barriers, and produce antimicrobial substances that inhibit different pathogenic microorganisms. For these reasons, zootechnical additives, made with Bacillus, are considered an alternative in the current livestock industry. The objective of this review was to show the probiotic potential in pig farming, poultry farming and calf rearing that Bacillus subtilis endospores have.

Currently, probiotics are used as animal growth promoters, since they improve the composition of the gastrointestinal microbiota and the efficiency in the use of food, stimulate the immune system and inhibit pathogenic microorganisms, without the use of antibiotics (Barros 2018Barros M. 2018. Among the probiotics used in livestock production are those made with of Bacillus spp. endospores (BioPlus 2B®, Biostart®, Toyocerin®, Liqualife®, Biosporin®, CenBiot®, Bactisubtil, Biosubtyl “Dalat” and Clostat®), with a probiotic effect on a wide category of zootechnical interest animals (Kizerwetter and Binek 2016Kizerwetter, S.M. & Binek, M. 2016. Due to the aforementioned, the objective of this review was to assess the results of researches on the probiotic potential of Bacillus subtilis endospores and their effect on pig farming, poultry farming and calf rearing.

Probiotic properties of bacillus subtilis endospores

 

Probiotics allow improvement in physiological indicators and stimulate action mechanisms to avoid side effects on animal products destined to human consumption. In the same way, they favor optimal growth of the animal, which provides good function of the intestinal mucosa, increased digestibility and the synthesis of vitamins; in addition to stimulating motility and the absence of diseases, important elements for production development (Sefer et al. 2015Sefer, D., Markovic, R., Nedeljkovic, T. In addition, they generate the stimulation of the specific immune response of the animal, and this allows the increase in immunoglobulin levels, which is a positive effect on growth and production (Kassa 2016Kassa, R. 2016. There are several action mechanisms of probiotics made with Bacillus strains, which favor the above.

1) Production of antimicrobial substances. They participate in the destruction of target cells through pore formation or inhibition of cell wall synthesis. In the case of bacteriocins, nisin, for example, forms a complex with the last cell wall precursor, thus inhibiting its biosynthesis in endospore-forming bacilli. Subsequently, it constitutes a complex that adds and incorporates peptides to form a pore in the bacterial membrane and generate cell death (Tao et al. 2018 Tao, M., Yutaka, S. & Le, L.G. 2018. "Dissect the mode of action of probiotics in affecting host-microbial interactions and immunity in food producing animals". Figure 1 describes the morphological changes that occur in a bacterial population after exposure to nisin.

2) Production of specific enzymes. Enzymes used as additives in animal feeding are abundant and varied. Pérez-Chabela et al. ISSN: 2448-7333. http://doi.org/10.24275/uam/izt/dcbs/hidro/2020v30n1/perez. report that among the enzymes produced by Bacillus spp in the vegetative phase are lytic enzymes (proteases, chitinases, cellulase, ß-1,3-glucanases, among others). Its use favors the synergistic action of these proteins on the most complex substrates present in foods. Generally, its use is aimed at improving the availability of polysaccharides, lipids and proteins, which are protected from digestive enzymes by impermeable structures of the plants cell wall, and also to degrade compounds that interfere with digestion, absorption and use of nutrients (Aftab and Bedford 2018Aftab, U. & Bedford, M.R. 2018. In recent years, the effective use of cellulases, xylanases and β-mannanases, as additives in diets for monogastrics, generated great interest from farmers (Alagawany et al. 2018Alagawany, M., Elnesr, S.S. & Farag, M.R. 2018. "The role of exogenous enzymes in promoting growth and improving nutrient digestibility in poultry". Iranian Journal of Veterinary Research, 19(3): 157-164, ISSN: 1728-1997. ).

Medina-Saavedra et al. (2017)Medina-Saavedra, T., Arroyo-Figueroa, G., Herrera-Méndez, C. & Mexicano-Santoyo, L. 2017. "Bacillus subtilis as a probiotic in poultry farming: relevant aspects in recent research". Abanico Veterinario, 7(3): 14-20, ISSN: 2448-6132. https://doi.org/10.21929/abavet2017.73.1. found that B. subtilis produces xylanase, which has a similar effect to antibiotics on the microbiota in the small intestine. The reduction in viscosity accelerates the intestinal transit speed. These authors reported that in the hydrolysis of arabinoxylans (AX), xylanase reduces the antinutrient effect, and thus increases its nutritional value.

Bedford (2018)Bedford, M.R. 2018. "The evolution and application of enzymes in the animal feed industry: the role of data interpretation data interpretation". British Poultry Science, 59(5): 486-493, ISSN: 1466-179. https://doi.org/10.1080/00071668.2018.1484074. added the enzyme xylanase to the diet for laying hens and observed adaptive changes in the microbiome of birds, in addition to verifying that the ability to degrade fibrous sources that are not hydrolyzed in the intestine of this species increased. The inclusion of β-mannanases in soybean-containing diets for broilers at the starter stage increased blood glucose content, anabolic hormone homeostasis, and amino acid digestibility (Caldas et al. 2018Caldas, J.V., Vignale, K., Boonsinchai, N., Wang, J., Putsakum, M., England, J.A. & Coon, C.N. 2018. "The effect of β -mananase on nutrient utilization and blood parameters in chicks fed diets containing soybean meal and guar gum". Poultry Science, 97(8): 2807-2817, ISSN: 0032-5791. https://doi.org/10.3382/ps/pey099. ). Likewise, the addition of this enzyme in diets composed of corn and soybean increased broilers yield, by reducing the content of galactomannans in the feed (Latham et al. 2018Latham, R.E., Williams, M.P., Walters, H.G., Carter, B. & Lee, J.T. 2018. "Efficacy of β -mannanase on broiler growth performance and energy utilization in the presence of increasing dietary galactomannan". The use of enzyme cocktails in laying hens improved nutrient retention and showed a tendency to increase enzyme activity in the intestine (Wen et al. 2012Wen, C., Wang, L.C., Zhou, Y.M., Jiang, Z.Y. & Wang, T. 2012. "Effect of enzyme preparation on egg production, nutrient retention, digestive enzyme activities and pancreatic enzyme messenger RNA expression of late-phase laying hens". Animal Feed Science and Technology, 172 (3-4): 180- 186, ISSN: 0377-8401. https://doi.org/10.1016/j.anifeedsci.2011.11.012.). El-Hack et al. (2017)El-Hack, M.E.A., Alagawany, M., Laudadio, V., Demauro, R. & Tufarelli, V. 2017. "Dietary inclusion of raw faba bean instead of soybean meal and enzyme supplementation in laying hens: Effect on performance and egg quality". Saudi Journal of Biological Sciences, 24(2): 276-285, ISSN: 1319-562X. https://doi.org/10.1016/j.sjbs.2015.05.009. pointed out that in this same category, the substitution of soybeans for beans (Vacia faba L.), supplemented with cellulases, xylanases, α-amylases and proteases, improved food efficiency, without affecting the quality and productivity of eggs.

3) Competence to prevent the adhesion of pathogens to epithelial cells. The mechanism of antipathogen competition is described as a process in which a bacterial species rigorously competes for adhesion to receptors in the gastrointestinal tract of an animal (figure 2) (Van et al. 2020Van, W., Deane, S. & Dicks, L. 2020. "Molecular insights into probiotic mechanisms of action employed against intestinal pathogenic bacteria". ISSN: 2448-7333. http://doi.org/10.24275/uam/izt/dcbs/hidro/2020v30n1/perez. reported some of the mechanisms used by Bacillus spp. to prevent the pathogens adhesion: 1) cell depolarization due to the formation of pores in the cell membrane, 2) inhibition of growth due to competition in adhesion sites and 3) inhibition of the expression of virulence genes.

4) Modulation of intestinal immunity. One of the characteristics that distinguish probiotics with Bacillus endospores is the ability to activate the immune system. The implementation of strategies for the care of the microbiota helps the host to maintain normal immune function through the expression of molecular patterns associated with metabolites derived from enzymes and antigens (figure 3). Therefore, the immune system heavily depends on the commensal microbiota for protection against invading pathogens (Tao et al. 2018 Tao, M., Yutaka, S. & Le, L.G. 2018. "Dissect the mode of action of probiotics in affecting host-microbial interactions and immunity in food producing animals". Veterinary Immunology and Immunopathology, 205: 35-48, ISSN: 1873-2534. https://doi.org/10.1016/j.vetimm.2018.10.004. ). In turn, probiotics have an immunomodulatory effect, which stimulates phagocytosis and the proliferation of immune cells (macrophages, monocytes and specialized cells, such as CD3, CD4 and CD8 T), in addition to the formation of antibodies (IgM and IgG) (Romero et al. 2013Romero, L. F., Parsons, C. M., Utterback, P. L., Plumstead, P. W. & Ravindran, V. 2013. In Cuba, there is the SUBTILPROBIO® probiotic product made with the strain of Bacillus subtilis subspecies subtilis (C-31, C-34 and E-44). The use of this product shows its in vitro probiotic properties (Milián et al. 2017Milián, G., Rondón, A.J., Pérez, M., Arteaga, F., Bocourt, R., Portilla, Y., Rodríguez, M., Pérez, Y., Beruvides, A. & Laurencio, M. 2017. "Methodology for the isolation, identification and selection of Bacillus spp. strains for the preparation of animal additives". Cuban Journal of Agricultural Science, 51(2): 197-207, ISSN: 2079-3480.).

The studies carried out by Milián (2009)Milián, G. 2009. Obtención de cultivos de Bacillus spp. y sus endosporas. Evaluación de su actividad probiótica en pollos (Gallus gallus domesticus). PhD Thesis, Instituto de Ciencia Animal, San José de las Lajas, Cuba, 100 p. show that the three strains (C-31, C-34 and E-44) have great capacity for growth and production of endospores, capable of inhibiting Gram-positive and Gram-negative microorganisms; in addition to showing sensitivity to a wide group of antibiotics and producing a group of specific enzymes (alkaline phosphatase, C4 esterase, C8 lipase esterase, C14 lipase, leucine arylamidase, naphthol-A-S-BI phosphohydrolase, α-glucosidase and β-glucosidase). This allows inferring that these strains show favorable ranges for their use as zootechnical additives, which constitute a promising alternative to the use of growth-promoting antibiotics.

5) Effect of Bacillus subtilis endospores in pigs. One of the main objectives of pig production today is to obtain the highest number of weaned piglets per sow in the year, healthy and of good weight. Among the nutritional strategies to improve sow yield, which is generally associated with better efficiency in nutrients use, is the utilization of additives. Among those used in sow feeding are probiotics, which show beneficial results related to milk production and quality (Rocha et al. 2018Rocha, V., Gobira, G., Andrade, T., Watanabe, P., Araújo, L., Gonçalves, M., Maciel, J., Martins, L., Bezerra, B. & Evangelista, J. 2018. Peet et al. (2020)Peet, S.C.M.C., Verheijen, R., Jørgensen, L. & Raff, L. 2020. "Effects of a mixture of Bacillus amyloliquefaciens and Bacillus subtilis on the performance of growing-_nishing pigs". Animal Feed Science and Technology, 261: 114409, ISSN: 0377-8401. https://doi.org/10.1016/j.anifeedsci.2020.114409. studied the effects of a probiotic supplement with Bacillus (mixture of spores of Bacillus amyloliquefaciens DSM 25840 and Bacillus subtilis DSM 32324) on the growth and health of fattening pigs. As a result of the study, they managed to improve the conversion rate and the average daily gain. Raudez and García (2020)Raudez, M.A.S. & García, W.M.O. 2020. Camoapa, Boaco, Nicaragua, p.66. evaluated the effectiveness of a probiotic with Lactobacillus lactis, Bacillus subtilis, Lactobacillus acidophilus, Bifidobacterium bifidum (PORCI-BIOTIC COMPLEX) for 28 d. These authors obtained favorable results for the animal health behavior variable, where the control group showed a greater number of respiratory and pulmonary diseases compared to the rest (PORCI-BIOTIC COMPLEX).

"Probiotic potential of Lactobacillus salivarius in animals of zootechnical interest"Cuban Journal of Agricultural Science, 54(2): 1-11, ISSN: 2079-3480. evaluated the probiotic effect of the biopreparations PROBIOLACTl®, SUBTILPROBIO® and their mixture on productive and health indicators of growing pigs. The evaluated biopreparations produced benefits in the animals, since they improved the eubiosis of the gastrointestinal tract, which contributed to improve (P<0.05) the live weight (27.15 kg/25.59 kg), the average daily gain of animals (408.65 g/445.27 g), weight gain (19.42 kg/16.36 kg) and feed conversion (2.44/2.90). In addition, the incidence of diarrhea decreased (8.57/67.14 %) in the treated animals. The results confirmed the probiotic potential of these biopreparations, when applied to pigs during the growth stage.

6) Effect of Bacillus subtilis endospores on birds. There are several researchers that show the positive response of the inclusion of zootechnical additives with a probiotic effect on the food for the poultry category, from probiotics with Bacillus endospores. Morales et al. (2020)Morales, P.J., Cortes, A.C., Gómez, G.V., Ávila, E.G., Arce, J. M., Del Río, J.C.G. 2020. "Efecto de un complejo multienzimático y un probiótico en gallinas de postura alimentadas con dietas sorgo-soya-canola". Revista Mexicana de Ciencias Pecuarias, 11(2): 369-379, ISSN: 2448-6698. https://doi.org/10.22319/rmcp.v11i2.4843. show this when they evaluated a multienzyme complex (proteases, amylases and xylanases) and a probiotic (Bacillus subtilis) in Bovans White hens. These authors confirmed an increase in egg weight with the addition of enzymes and probiotics. However, for the humoral immunity, cholesterol, LDL and HDL variables, there were not differences (P>0.05) between treatments.

Cuban Journal of Agricultural Science, 53(3): 241-248, ISSN: 2079-3480. evaluated the zootechnical additive SUBTILPROBIO® in laying hens of Leghorn breed L33. To determine the probiotic effect, they measured: live weight, intake, conversion, total egg production, cracked eggs and total disqualified eggs, as well as mortality, death by pecking or cannibalism and viability. The results showed improvement of the indicators live weight (1640.0 g/1585.0 g), intake (10780 kg DM), conversion (1.92/2.10), egg production (15 540/15 397), craked eggs (1092/1114) and disqualified (69/76) for P<0.01 with respect to the control group. The indicators mortality (1/4), viability (99.6/98.6 %) and death by pecking/cannibalism (1/2) did not show differences between treatments.

Cuban Journal of Agricultural Science, 5(1): 67-75, ISSN: 2079-3480., when evaluating the zootechnical additive SUBTILPROBIO® E-44 in productive and health indicators in Heavy Pure Breed birds, recently reported that the productive indicators live weight, uniformity and conversion showed significant values for (P<0.001) with respect to the control group and the standard for the breed under study. The indicators mortality (3.8/8) and viability (96.2/92 %) showed differences with respect to the control for P<0.01.

"Efecto de un complejo multienzimático y un probiótico en gallinas de postura alimentadas con dietas sorgo-soya-canola". Revista Mexicana de Ciencias Pecuarias, 11(2): 369-379, ISSN: 2448-6698. https://doi.org/10.22319/rmcp.v11i2.4843. showed that when they evaluated a multienzyme complex, composed of amylases, proteases and xylanases, and the probiotic with Bacillus subtilis spores, in sorghum-soybean-rapeseed diets, the results of productive yield showed differences (P<0.05) in egg weight. These authors reported that lower weight was obtained with the treatment (60.0/58.9/59.2), but with the addition of enzymes and probiotics, an increase was shown. For the variables cholesterol (100.2/109.7/122.9 mg/dL), LDL (13.9/17.3/16.2 mg/dL) and HDL (30.0/37.0/35.7 mg/dL) there were not differences (P>0.05) between treatments.

Revista de Ciencias Agropecuarias ALLPA, 2(4): 1-17, ISSN: 2600-5883. https://publicacionescd.uleam.edu.ec/index.php//allpa/isue/view/19. evaluated a Bacillus subtilis probiotic in Cobb 500 broiler category. The measured indicators were productive, such as weight gain, feed conversion rate, American efficiency factor, carcass weight, mortality, changes in villi and pathological analysis. The mentioned authors showed that the probiotic Bacillus subtilis has a positive effect on the productive parameters.

Regarding the use of probiotics in poultry farming, there are infinite studies that report and show the effectiveness of biopreparations with Bacillus spp. endospores, a criterion that is refuted by the Engineering your feed solutions (ORFFA). This entity ensures that sporulated probiotics are the most natural option to produce healthy birds, highly effective and very competitive, if used in poultry production, so that their productive yields were optimized (ORFFA 2021ORFFA. 2021. 7) Effect of Bacillus subtilis endospores on calves. In the evaluation studies of the zootechnical additive SUBTILPROBIO®, in the category of lactating calves from Siboney de Cuba breed, Hernández (2012)Hernández, Y. 2012. Elaboración del SUBTILPROBIO® en terneros lactantes de la Recría "Los Quinientos". Diploma Thesis SUM “Jesús Herrera” Pedro Betancourt. Universidad de Matanzas, Cuba, p.90. observed that the calves witch intake the zootechnical additive had a live weight higher (probiotic GT 169/Control group 110 kg) upon transfer to the development unit. In this case, the health indicators, incidence of diarrhea (1/7) and pneumonia (-/2), and mortality (-/4) were measured for P≤0.05.

Silva (2013)Silva, Y. 2013. Efecto probiótico de un biopreparado de Bacillus subtilis C-31 en terneros lactantes. Diploma Thesis. Universidad de Matanzas, Cuba. p 100. showed that by supplying the probiotic biopreparation SUBTILPROBIO® C-31 in lactating calves from Siboney de Cuba breed for 90 days, an increase in the productive weight indicator (74.6/58.3 kg) is achieved with respect to the control for P≤0.05.

Conclusions

 

Bacillus subtilis is a bacterium with a probiotic effect which is in the digestive tract of zootechnical interest animals and in other environments. The researches carried out to date show the in vitro and in vivo potentialities of this bacterium and its endospores.

En la actualidad, las producciones avícolas, porcinas y bovinas constituyen las ramas de la producción animal de mayor importancia en el mundo, lo que contribuye en estos momentos a la satisfacción de las necesidades proteicas de la población mundial. Se pronostica el uso de probióticos, prebióticos y simbióticos con el propósito de utilizarlos como promotores del crecimiento animal. Entre las especies que más se utilizan para la elaboración de estos productos, están las del género Bacillus. Una de ellas es Bacillus subtilis, una bacteria capaz de estimular el sistema inmunológico, producir enzimas, resistir las barreras gástricas de pH ácido, y producir sustancias antimicrobianas que inhiben diferentes microorganismos patógenos. Por estas razones, los aditivos zootécnicos, elaborados con Bacillus, se consideran una alternativa en la industria pecuaria actual. Esta reseña tiene como objetivo demostrar el potencial probiótico en la porcinocultura, avicultura y crianza de terneros que tienen las endosporas de Bacillus subtilis.

En la actualidad, se utilizan probióticos como promotores del crecimiento animal, pues mejoran la composición de la microbiota gastrointestinal y la eficiencia en el uso de los alimentos, estimulan el sistema inmune e inhiben microorganismos patógenos, sin la utilización de antibióticos (Barros 2018Barros M. 2018. Por lo antes referido, el objetivo de esta reseña fue valorar los resultados de las investigaciones acerca del potencial probiótico de las endosporas de Bacillus subtilis y su efecto en la porcinocultura, avicultura y crianza de terneros.

Propiedades probióticas de las endosporas de bacillus subtilis

 

Los probióticos permiten la mejora en los indicadores fisiológicos y estimulan los mecanismos de acción para evitar efectos secundarios en productos de origen animal destinados al consumo humano. Del mismo modo, favorecen un óptimo crecimiento del animal, lo que proporciona buena función de la mucosa intestinal, aumento de la digestibilidad y la síntesis de vitaminas; además de estimular la motilidad y la ausencia de enfermedades, elementos importantes para el desarrollo de la producción (Sefer et al. 2015Sefer, D., Markovic, R., Nedeljkovic, T. J., Petrujkic, B., Radulovic, S. & Grdovic S. 2015. "The application of biotechnology in animal nutrition". Además, generan la estimulación de la respuesta inmune específica del animal, y esto permite el incremento en los valores de inmunoglobulina, lo que se traduce en un efecto positivo en el crecimiento y la producción (Kassa 2016Kassa, R. 2016. Existen varios mecanismos de acción de los probióticos elaborados con cepas de Bacillus, que favorecen lo antes mencionado.

1) Producción de sustancias antimicrobianas. Participan en la destrucción de las células diana mediante la formación de poros o la inhibición de la síntesis de la pared celular. En el caso de las bacteriocinas, la nisina, por ejemplo, forma un complejo con el último precursor de la pared celular, y de esta forma se inhibe su biosíntesis en bacilos formadores de endosporas. Posteriormente, constituye un complejo que agrega e incorpora péptidos para formar un poro en la membrana bacteriana y generar la muerte celular (Tao et al. 2018 Tao, M., Yutaka, S. & Le, L.G. 2018. En la figura 1 se describen los cambios morfológicos que ocurren en una población bacteriana tras la exposición a la nisina.

2) Producción de enzimas específicas. Las enzimas que se utilizan como aditivos en la alimentación animal son abundantes y variadas. Pérez-Chabela et al. ISSN: 2448-7333. http://doi.org/10.24275/uam/izt/dcbs/hidro/2020v30n1/perez. refieren que entre las enzimas que produce Bacillus spp en la fase vegetativa se encuentran las enzimas líticas (proteasas, quitinasas, celulasa, ß-1,3-glucanasas, entre otras). Su utilización favorece la acción sinérgica de estas proteínas en los sustratos más complejos presentes en los alimentos. Generalmente, su utilización se dirige a mejorar la disponibilidad de polisacáridos, lípidos y proteínas, que se encuentran protegidos de las enzimas digestivas por estructuras impermeables de la pared celular de las plantas, y también a degradar compuestos que interfieren en la digestión, absorción y utilización de nutrientes (Aftab y Bedford 2018Aftab, U. & Bedford, M.R. 2018. En los últimos años, el uso efectivo de las celulasas, xilanasas y β-mananasas, como aditivos en las dietas para monogástricos, generó gran interés por parte de los productores (Alagawany et al. 2018Alagawany, M., Elnesr, S.S. & Farag, M.R. 2018. "The role of exogenous enzymes in promoting growth and improving nutrient digestibility in poultry". Iranian Journal of Veterinary Research, 19(3): 157-164, ISSN: 1728-1997. ).

Medina-Saavereda et al. (2017)Medina-Saavedra, T., Arroyo-Figueroa, G., Herrera-Méndez, C. & Mexicano-Santoyo, L. 2017. "Bacillus subtilis as a probiotic in poultry farming: relevant aspects in recent research". Abanico Veterinario, 7(3): 14-20, ISSN: 2448-6132. https://doi.org/10.21929/abavet2017.73.1. encontraron que B. subtilis produce xilanasa, la que tiene un efecto similar a los antibióticos en la microbiota a nivel del intestino delgado. La reducción de la viscosidad acelera la velocidad de tránsito intestinal. Estos autores refirieron que en la hidrólisis de los arabinoxilanos (AX), la xilanasa permite reducir el efecto antinutriente, y aumentar así su valor alimenticio.

British Poultry Science, 59(5): 486-493, ISSN: 1466-179. https://doi.org/10.1080/00071668.2018.1484074. adicionó en la dieta para gallinas ponedoras la enzima xilanasa y observó cambios adaptativos en el microbioma de las aves, además de comprobar que se incrementó la capacidad para degradar las fuentes fibrosas que no se hidrolizan en el intestino de esta especie. La inclusión de β-mananasas en dietas que contenían soya para pollos de ceba en la etapa de inicio aumentó el contenido de glucosa en sangre, la homeostasis de las hormonas anabólicas y la digestibilidad de los aminoácidos (Caldas et al. 2018Caldas, J.V., Vignale, K., Boonsinchai, N., Wang, J., Putsakum, M., England, J.A. & Coon, C.N. 2018. Asimismo, la adición de esta enzima en dietas compuestas por maíz y soya aumentó el rendimiento de los pollos de ceba, al disminuir el contenido de galactomananos del pienso (Latham et al. 2018Latham, R.E., Williams, M.P., Walters, H.G., Carter, B. & Lee, J.T. 2018. "Efficacy of β -mannanase on broiler growth performance and energy utilization in the presence of increasing dietary galactomannan". Poultry Science, 97(2): 549-556, ISSN: 0032-5791. https://doi.org/10.3382/ps/pex309. ).

El uso de cócteles enzimáticos en gallinas ponedoras mejoró la retención de nutrientes y mostró tendencia al incremento de la actividad de las enzimas en el intestino (Wen et al. 2012Wen, C., Wang, L.C., Zhou, Y.M., Jiang, Z.Y. & Wang, T. 2012. "Effect of enzyme preparation on egg production, nutrient retention, digestive enzyme activities and pancreatic enzyme messenger RNA expression of late-phase laying hens". (2017)El-Hack, M.E.A., Alagawany, M., Laudadio, V., Demauro, R. & Tufarelli, V. 2017. "Dietary inclusion of raw faba bean instead of soybean meal and enzyme supplementation in laying hens: Effect on performance and egg quality". Saudi Journal of Biological Sciences, 24(2): 276-285, ISSN: 1319-562X. https://doi.org/10.1016/j.sjbs.2015.05.009. señalaron que en esta misma categoría, la sustitución de soya por fabas (Vacia faba L.), suplementada con celulasas, xilanasas, α-amilasas y proteasas, mejoró la eficiencia alimentaria, sin afectar la calidad y la productividad de los huevos.

3) Competencia para evitar la adhesión de patógenos a las células epiteliales. El mecanismo de competencia antipatógenos se describe como un proceso en el que una especie bacteriana compite rigurosamente por la adhesión a los receptores en el tracto gastrointestinal de un animal (figura 2) (Van et al. 2020Van, W., Deane, S. & Dicks, L. 2020. ISSN: 2448-7333. http://doi.org/10.24275/uam/izt/dcbs/hidro/2020v30n1/perez. refirieron algunos de los mecanismos utilizados por Bacillus spp. para evitar la adhesión de patógenos: 1) despolarización celular por formación de poros en la membrana celular, 2) inhibición del crecimiento por competencia en sitios de adhesión y 3) inhibición de la expresión de genes de virulencia.

4) Modulación de la inmunidad intestinal. Una de las características que distingue a los probióticos con endosporas de Bacillus es la capacidad de activar el sistema inmunológico. La implementación de estrategias para el cuidado de la microbiota ayuda al huésped a mantener una función inmunológica normal mediante la expresión de patrones moleculares asociados a metabolitos derivados de enzimas y antígenos (figura 3). Por lo tanto, el sistema inmunológico depende, en gran medida, de la microbiota comensal para la protección contra patógenos invasores (Tao et al. 2018 Tao, M., Yutaka, S. & Le, L.G. 2018. "Dissect the mode of action of probiotics in affecting host-microbial interactions and immunity in food producing animals". A su vez, los probióticos tienen un efecto inmunomodulador, que estimula la fagocitosis y la proliferación de células inmunes (macrófagos, monocitos y células especializadas, como CD3, CD4 y CD8 T), además de la formación de anticuerpos (IgM y IgG) (Romero et al. 2013Romero, L. En Cuba, se cuenta con el producto probiótico SUBTILPROBIO® elaborado con la cepa de Bacillus subtilis subespecie subtilis (C-31, C-34 y E-44). El uso de este producto evidencia sus propiedades probióticas in vitro (Milián et al. 2017Milián, G., Rondón, A.J., Pérez, M., Arteaga, F., Bocourt, R., Portilla, Y., Rodríguez, M., Pérez, Y., Beruvides, A. & Laurencio, M. 2017. "Methodology for the isolation, identification and selection of Bacillus spp. strains for the preparation of animal additives". Cuban Journal of Agricultural Science, 51(2): 197-207, ISSN: 2079-3480.).

Los estudios realizados por Milián (2009)Milián, G. 2009. Obtención de cultivos de Bacillus spp. y sus endosporas. Evaluación de su actividad probiótica en pollos (Gallus gallus domesticus). PhD Thesis, Instituto de Ciencia Animal, San José de las Lajas, Cuba, 100 p. muestran que las tres cepas (C-31, C-34 y E-44) tienen gran capacidad de crecimiento y producción de endosporas, capaces de inhibir microorganismos Gram positivos y Gram negativos; además de mostrar sensibilidad a un amplio grupo de antibióticos y producir un grupo de enzimas específicas (fosfatasa alcalina, esterasa C4, esterasa lipasa C8, lipasa C14, leucina arilamidasa, naftol-A-S-BI fosfohidrolasa, α-glucosidasa y β-glucosidasa). Esto permite inferir que dichas cepas muestran rangos favorables para su utilización como aditivos zootécnicos, que constituyen una alternativa prometedora ante el uso de antibióticos promotores del crecimiento.

5) Efecto de las endosporas de Bacillus subtilis en cerdos. Uno de los principales objetivos de la producción porcina en la actualidad es obtener el mayor número de lechones destetados por cerda en el año, saludables y de buen peso. Entre las estrategias nutricionales para mejorar el rendimiento de las cerdas, que se asocia generalmente con una mejor eficiencia en la utilización de nutrientes, se encuentra el uso de aditivos. Entre los manejados en la alimentación de cerdas, se encuentran los probióticos, que muestran resultados beneficiosos, relacionados con la producción y calidad de la leche (Rocha et al. 2018Rocha, V., Gobira, G., Andrade, T., Watanabe, P., Araújo, L., Gonçalves, M., Maciel, J., Martins, L., Bezerra, B. & Evangelista, J. 2018. Efeito da suplementação de levedura em matrizes suínas no terço final da gestação e na lactação em clima tropical sobre o desempenho da leitegada. Anais do IX Forum Internacional de Suinocultura. Animal Feed Science and Technology, 261: 114409, ISSN: 0377-8401. https://doi.org/10.1016/j.anifeedsci.2020.114409. estudiaron los efectos de un suplemento probiótico con Bacillus (mezcla de esporas de Bacillus amyloliquefaciens DSM 25840 y Bacillus subtilis DSM 32324) en el crecimiento y la salud de cerdos de engorde. Como resultado del estudio, lograron mejorar el índice de conversión y la ganancia media diaria. Raudez y García (2020)Raudez, M.A.S. & García, W.M.O. 2020. Evaluación del uso de probióticos en la producción de cerdos post-destete de genética Topigs Norsvin en la Finca El Porvenir, Municipio de Mulukukú, departamento de la RACCN. Trabajo de Tesis para optar por el título de Ingeniero Agrónomo. Camoapa, Boaco, Nicaragua, p.66. evaluaron la efectividad de un probiótico con Lactobacillus lactis, Bacillus subtilis, Lactobacillus acidophilus, Bifidobacterium bifidum (PORCI-BIOTIC COMPLEX) durante 28 d. Estos autores obtuvieron resultados favorables para la variable comportamiento zoosanitario, donde el grupo testigo presentó mayor cantidad de enfermedades respiratorias y pulmonares en comparación con el resto (PORCI-BIOTIC COMPLEX).

"Probiotic potential of Lactobacillus salivarius in animals of zootechnical interest"Cuban Journal of Agricultural Science, 54(2): 1-11, ISSN: 2079-3480. evaluaron el efecto probiótico de los biopreparados PROBIOLACTlL®, SUBTILPROBIO® y su mezcla en indicadores productivos y de salud de cerdos en crecimiento. Los biopreparados evaluados produjeron beneficios en los animales, ya que mejoraron la eubiosis del tracto gastrointestinal, lo que contribuyó a mejorar (P<0.05) el peso vivo (27.15 kg/25.59 kg), la ganancia media diaria de los animales (408.65 g/445.27g), el incremento de peso (19.42 kg/16.36 kg) y la conversión alimentaria (2.44/2.90). Además, disminuyeron la incidencia de diarreas (8.57/67.14 %) en los animales tratados. Los resultados confirmaron el potencial probiótico que tienen estos biopreparados, cuando se aplican a cerdos durante la etapa de crecimiento.

6) Efecto de las endosporas de Bacillus subtilis en aves. Diversas son las investigaciones que demuestran la respuesta positiva de la inclusión de aditivos zootécnicos con efecto probiótico en el alimento para la categoría avícola, a partir de probióticos con endosporas de Bacillus. Morales et al. (2020)Morales, P.J., Cortes, A.C., Gómez, G.V., Ávila, E.G., Arce, J. M., Del Río, J.C.G. 2020. "Efecto de un complejo multienzimático y un probiótico en gallinas de postura alimentadas con dietas sorgo-soya-canola". Revista Mexicana de Ciencias Pecuarias, 11(2): 369-379, ISSN: 2448-6698. https://doi.org/10.22319/rmcp.v11i2.4843. así lo demuestran, cuando evaluaron un complejo multienzimático (proteasas, amilasas y xilanasas) y un probiótico (Bacillus subtilis) en gallinas Bovans White. Estos autores comprobaron incremento en el peso de huevo con la adición de enzimas y probióticos. Sin embargo, para las variables inmunidad humoral, colesterol, LDL y HDL, no se observaron diferencias (P>0.05) entre tratamientos.

Poultry Science, 96(1): 74-82, ISSN: 0032-5791. https://doi.org/10.3382/ps/pew246. informaron mejoras en el aumento de peso y la tasa de conversión alimentaria en machos Arbor Acres de un día de nacidos, cuando suplementaron dietas basales con Bacillus subtilis mbJ (BSfmbJ) en dosis de 2, 3 y 4 x 1010 ufc/kg, sin usar promotores del crecimiento.

Milián et al. (2019)Milián, G.F., Rondón, A.J.C., Pérez, M.Q., Martínez, Y., Boucourt, R., Rodríguez, M.O., Beruvides, A. & Portilla, Y. 2019. "Stability of the zootechnical additives SUBTILPROBIO® C-31, C-34 and E-44 under different temperature conditions". Cuban Journal of Agricultural Science, 53(3): 241-248, ISSN: 2079-3480. evaluaron el aditivo zootécnico SUBTILPROBIO® en gallinas ponedoras de la raza Leghorn Línea L33. Para determinar el efecto probiótico midieron: peso vivo, consumo, conversión, producción total de huevos, huevos cascados y totales de huevos descalificados, así como la mortalidad, muerte por picaje o canibalismo y viabilidad. Los resultados mostraron mejora de los indicadores peso vivo (1640.0 g/1585.0 g), consumo (10780 kg MS), conversión (1.92/2.10), producción de huevos (15 540/15 397), huevos cascados (1092/1114) y descalificados (69/76) para P<0.01 con respecto al grupo control. Los indicadores mortalidad (1/4), viabilidad (99.6/98.6 %) y muerte por picaje/canibalismo (1/2) no tuvieron diferencias entre los tratamientos.

"Evaluation of the zootechnical additive SUBTILPROBIO® E-44 in productive and health indicators in heavy pure breeds birds under production conditions". Cuban Journal of Agricultural Science, 5(1): 67-75, ISSN: 2079-3480., al evaluar el aditivo zootécnico SUBTILPROBIO® E-44 en indicadores productivos y de salud en aves Líneas Puras Pesadas, informaron recientemente que los indicadores productivos peso vivo, uniformidad y conversión mostraron valores significativos para (P<0.001) con respecto al grupo control y al estándar para la línea en estudio. Los indicadores mortalidad (3.8/8) y viabilidad (96.2/92 %) dejaron ver diferencias con respecto al control para P<0.01.

Estudios realizados por Morales et al. (2020)Morales, P.J., Cortes, A.C., Gómez, G.V., Ávila, E.G., Arce, J. M., Del Río, J.C.G. 2020. "Efecto de un complejo multienzimático y un probiótico en gallinas de postura alimentadas con dietas sorgo-soya-canola". Revista Mexicana de Ciencias Pecuarias, 11(2): 369-379, ISSN: 2448-6698. https://doi.org/10.22319/rmcp.v11i2.4843. demostraron que cuando evaluaron un complejo multienzimático, compuesto de amilasas, proteasas y xilanasas, y el probiótico con esporas de Bacillus subtilis, en dietas de sorgo-soya-canola, los resultados de rendimiento productivo mostraron diferencias (P<0.05) en el peso de huevo. Estos autores refirieron que se obtuvo menor peso con el tratamiento (60.0/58.9/59.2), pero con la adición de enzimas y probióticos, se mostró incremento. Para las variables colesterol (100.2/109.7/122.9 mg/dL), LDL (13.9/17.3/16.2 mg/dL) y HDL (30.0/37.0/35.7 mg/dL) no se observaron diferencias (P>0.05) entre tratamientos.

Cuban Journal of Agricultural Science, 49(4): 497-502, ISSN: 2079-3480., al evaluar una mezcla probiótica de dos aditivos zootécnicos (PROBIOLACTIL® C65 y SUBTILPROBIO® E-44) con respecto al estándar en aves de Línea Pura Pesada B4 durante cinco semanas, obtuvieron resultados positivos en cuanto al incremento de peso desde la tercera semana de inclusión de los biopreparados (793, 1249 y 1587g). "Probiotic potential of Lactobacillus salivarius in animals of zootechnical interest"Cuban Journal of Agricultural Science, 54(2): 1-11, ISSN: 2079-3480., cuando refieren la utilización de mezclas de microorganismos en biopreparados destinados a la producción animal.

Los indicadores medidos fueron de tipo productivo, como ganancia de peso, índice de conversión alimentaria, factor de eficiencia americana, peso de la canal, mortalidad, cambios en las vellosidades y análisis patológico. Los autores citados demostraron que el probiótico Bacillus subtilis tiene un efecto positivo en los indicadores productivos.

Sin lugar a dudas, en la temática del uso de probióticos en la avicultura, son infinitos los estudios que informan y demuestran la efectividad de los biopreparados con endosporas de Bacillus spp., criterio que se refuta por la Engineering your feed solutions (ORFFA). 7) Efecto de las endosporas de Bacillus subtilis en terneros. En los estudios de la evaluación del aditivo zootécnico SUBTILPROBIO®, en la categoría de terneros lactantes de la raza Siboney de Cuba, Hernández (2012)Hernández, Y. 2012. Elaboración del SUBTILPROBIO® en terneros lactantes de la Recría "Los Quinientos". Diploma Thesis SUM “Jesús Herrera” Pedro Betancourt. Universidad de Matanzas, Cuba, p.90. observó que los terneros que consumieron el aditivo zootécnico tuvieron mayor peso vivo (GT probiótico 169/Grupo control 110 kg) a su traslado para la unidad de desarrollo. En este caso, se midieron los indicadores de salud, incidencia de diarreas (1/7) y de neumonía (-/2) y mortalidad (-/4) para P≤0.05.

Silva (2013)Silva, Y. 2013. Efecto probiótico de un biopreparado de Bacillus subtilis C-31 en terneros lactantes. Diploma Thesis. Universidad de Matanzas, Cuba. p 100. demostró que al suministrar el biopreparado probiótico SUBTILPROBIO® C-31 en terneros lactantes de la raza Siboney de Cuba durante 90 días, se logra un incremento en el indicador productivo peso (74.6/58.3 kg) con respecto al control para P≤0.05.

Conclusiones

 

Bacillus subtilis es una bacteria con efecto probiótico que se encuentra en el tracto digestivo de los animales de interés zootécnico y en otros ambientes. Las investigaciones realizadas hasta la fecha demuestran las potencialidades in vitro e in vivo de esta bacteria y sus endosporas.
